Durian (Durio zibethinus Murr.) is one of tropic fruits that has varian taste depending on the places their production. Nias Island, well known as durian production place in North Sumatera. The information about it’s was less studied and now day we need to analyze variant especially Island. aim this research genetic diversity Island with Random Amplified Polymorphic DNA (RAPD). Ten shoots collected from each 5 regencies, which were: Selatan, Barat, Utara, Kota, dan Induk. Durian’s been isolated by using Cetyl Trimethyl Ammonium Bromide (CTAB) method amplified six RAPD primers namely: OPA 01, 03, 07, 10, 05, OPN 06. band were scored translated biner data, then analyzed Numerical Taxonomy Multivariate System (NTSys) clustered Unweighted Pair Group Method With Arithme Average (UPGMA) method. Isolated showed a clear unsmeared band. size around 221-2855bp 100% polymorphism. dendogram constructed 50 accession Nias’s 0.51-0.59 similarity value population. All similiar 67% B2 (Nias Barat) farthest distance all accession.
